BioSpec

Some up-front notes:

* All supplies and accessories are located in the drawers near the spec in room 9.

* Under # 9 Utilities on the Spec monitor, one should run the performance check and baseline correction ~ every 2 months. See page 3-1 Daily Maintenance.

* The reference cell is in the back…..the sample cell is in the front.

* You can choose to run the Spec via the keyboard/monitor on the Spec or after initializing the Spec, you can use the UVPC software on the computer. You may start and run the Spec without the PC, BUT if you choose to control the Spec with the PC, then decide to switch back to the Spec controlling itself, you will have to shut down the Spec and restart.

* In the scanning mode, the Spec scans from the long to short wavelengths (ROYGBIVUV).

Figure 1
Figure 1. The Spec. See pages 2-2 > 2-6 for other external components. A – the sample compartment with cell holders shown at the far right. A single screw in the center holds the dual cell holder. We have three 10mm light path cell holders: regular, mini and temperature controlled. Two knobs at the bottom of A are used to release the sample compartment cover when you use the flow cells. B – The LCD monitor. The mode selection screen appears here as well as all graphs and data tables. C – The keyboard. This is used to select the MODE under which you want to operate and to enter numerical values (See Fig. 2).

2. The Spec will power up and carry out initialization (See Fig. 4). Then the MODE screen pops up (See Fig. 5). You can choose to use the keyboard to run the Spec [do nothing] OR you may use the computer to run the Spec. In the latter case, turn on the computer and double-click on the UVPC icon. Then hand over control of the Spec to the computer by pressing F4 [PC CTRL]. The computer allows you to do more processing and printing than the keyboard mode.
